Semaglutide - long-acting GLP-1 receptor agonist (Ozempic / Wegovy / Rybelsus) that reshaped weight management and type-2 diabetes care. Pictured is the lipid-modified Lys26 side-chain - the C18 fatty diacid plus γ-Glu/OEG2 spacer that gives semaglutide its multi-day half-life via albumin binding. On 28 January 2025 the FDA approved an additional Ozempic indication to reduce kidney-disease progression and cardiovascular death (FLOW: 24% RR reduction). In early 2026 the FDA cleared the Wegovy oral tablet (OASIS-4 Phase-3: 16.6% mean weight loss at 64 weeks vs 2.7% on placebo) - the first oral GLP-1 for weight loss and Novo Nordisk's answer to needle-averse patients and the pill-based competitors now entering the class.
